Ignore what the characters have in common. If Capcom and SNK can work together to make Capcom vs. SNK, why can't other companies work together. Anyways, an obvious proposed list is as follows:
(NOTE:Some obscure characters like Falcon and Ness may seem insignificant to us, but are hugely popular in Japan. Taking this to note, we are likely to see:
Mario
Luigi
Peach
Yoshi (Boshi? Come on)
(These four are staple characters)
Kirby
(HAL Labs made the game)
Ness
(Mother 1 and 2 were hugely popular, so is the now defunct Mother 64)
Pikachu, Satoshi, Kasumi
(Give me a break, you realize this must appeal to the masses)
Donkey Kong
(They have a CCG in Japan on the 3D Fox Family TV Show)
Bowser & Ganondorf, possibly Wario
(The old one featured no villans)
Link, possibly Young after Majora's Mask
Zelda is a very slim chance
Samus returns with a revamp, based on the upcoming GameCube version
Little Mac may see the day, being that Ready 2 Rumble is so popular and a remake seems likely
MewTwo or Pichu also seem like strong possibilities. (In fact in the current Issue of the Japanese Monthly Pokemon Card Game Charizard Base Set is still the highest value card, some Neo's are on there, too, though)
The bit characters like Birdo and Waluigi might get a break in, if Camelot Software Planning designs the new one instead of HAL, or maybe they won't have to.
Megaman seems very unlikely, considering his creator is forsaking him after X5, Xtreme, and EXE after seeing a quote to the tune of "We have run out of ideas"
Sonic is actually possible being the NCL and Sega have had talks together about a partnership
Crash is just annoying
Kid Icarus might be a hidden character
Bomberman would be nice, considering that Hudson designed Mario Party and have good NCL relations.
I imagine Cameos of various enemies like Goombas and Lakitus plus Pokemon Appearances.
The real question is about who will develop it.
Can HAL strike gold again?
Could HUDSON do it?
Camelot Software Planning is becoming very popular.
Capcom has just finished porting MM Legends, could they take the job?
Saffire, who made Xena:Talisman of Fate wouldn't do half bad.

Yeah, I got tickets from Saffire, a programming company. They had the SGAG (Saffire Gaming Advisory Group) and now is defunct. But I got tickets from them for free. Yay!

Deezer, did you go to E3? I read somewhere about the Gamecube turning the 128 Marios 2D. I also played PMS at E3, is it the same effect, or more like Jet Grind Radio? Either is cool. BTW, this is my new name, I previously was known as JayGee1

I like my Level 50 Golfer Kid in Mario Golf. Besides that I take a liking to Yoshi (Even is SSB) BTW, I have a hunch Bowser's gonna be in SSB2.

I change my answer. You'd be hard pressed to find it, but a game called Hotel Mario was once on the CD-i, check out the instruction booklet I typed up on it. A stinker if I ever saw one, put breifly the koopa kids cheat in the game and it gets repetitive after the first level.
BTW, I like Super Mario Land, on trips I beat it every time, it's fun and easy.
