Has there ever been a time when you beat something that looked seemingly impossible in a Mario game? Here are some of mine.

Beating attack 15 in Mario Party Advance.
Beating 100 wins in endurance alley.
Hiting a Grand Slam and getting Mario to a Superstar in MSSB.
Doing the 16 star completion in SM64.
Getting every Blue coin in SMS.
Getting a gold portrait of every ghost in Luigi's Mansion!
Getting past Bowser in SMB 8-4 while small in hard mode.
Getting every character in MG:TT.
Getting 100 in every level in YI!!!!!

Nintendo,Mario,and Bomberman pwns. Sony,Microsoft,and Halo are n00bs.

Getting killed five times in a row without getting at single point in SMB. Hey, *I* thought it was impossible.

Beating Bowser in Tetris Attack standard mode, the FIRST time he appears, and watching him call Yoshi a donkey.

Beating 15-minute Melee is easy with Donkey Kong.

Defeating Bonetail and getting through the Pit of 100 Trials. Poor partners were on freakin' life support by the time I won the battle.

Kelso: Guys this is a game of Cat & Mouse between me & Hyde. I''m gonna make him admit he''s been going out with Jackie behind my back.
Fez: Guys, what happens in Cat & Mouse if the cat is retarded?
As a game that requires six friends, an HDTV, and skill, I can see why the majority of TMK is going to hate on it hard.

For me, it was getting "Wario's Hamburger" in MP4.

I hope you don't mind but I'm going to talk about something that doesn't really involve beating things (so I'm going along with just the title).  Paper Mario 2 had a ton of great funny moments for me.  Such as playing the SMB levels with Bowser, reaching the entrance of the Boggly Tree before the bomb went off only to see two guys wasting more time and freaking me out, "I love you" x 100, the easy but very funny boss Doopliss, and probably most of all the entire level of Glitz Pit.

Now for accomplishments.  Mario Sunshine had a ton of hard bonus levels that felt good to beat (I still have to beat a few more next time I rent this).  Break the Targets in SSb was another toughie before.  The Lost Levels drove me insane for a long time.  I played SSBM forever to unlock secret characters and my mom got worried when she heard how long I left the game on.  SMW and Special... you know what I mean.  Yikes.  100 Super Jumps in SMRPG.  Yowza.  But after you beat all this stuff you feel pretty good (and stressed out).

In Super Mario 128, I had a lot of trouble trying to unlock Donkey Kong and E. Gadd, and also finding the hammer suit.  Oh sorry... I keep forgetting that I went ahead in time while you guys haven't got there yet.

And let us not be weary in well doing: for in due season we shall reap, if we faint not.

The cake is a lie, your base belongs to us, keyboard cat will play you off as you fall out of the bus.
